You have something blocking it.
Try plunging some more. Use a good power plunger. It that fails try putting a quart of liquid plumber in it and let it set a couple hours.
Are you missing anything in the bathroom? A comb, toothbrush, makeup? There things get flushed and hang up in the U of the toilet itself. That would mean removing the toilet.
I really think from your description a real good plunger will work.

try pouring a bucket of water down it. the pressure should help move anything that is stuck to go on down the pipes.

Many reasons/solutions:
Try to pour a large bucket of water forcefully.
Try a rotorouter.
May be a tree root has grown inside the drain that connects your line to the sewer. This is a big job to get rid off. This means digging in your side/front yards.
The vent pipe is blocked. Every house has one, to facilitate water drain into the sewage. Inspect. Clue: If other toilets in the house do the same thing, it is the vent pipe problem.
Other than that, throw you arms up in the air, and call a plummer. Take a second mortgage to pay his fees.
We experience this on occasion. The roots from the tree out by the street grow into the lines. How old is your house? Old lines were made with something called orange gorp, which is just compressed tar paper ( if you can believe that). You probably have some kind of clog in your main line. It should be visible some where at the front of your house and the cap should just come off, it's like an overflow. Locate that, uncap it, run water or flush toiler to see if it overflows. This can help you detect where the clog is. It can be rootered out. Some times the roof vent for the toilets also get clogged with leaves and debri. Check those as well to make sure thay have air flow.

Also, you may want to check your roof vent pipe. We've had this problem with my mom where the vent was clogging (like putting your finger over the hole at the top of a straw). This site also mentions it:
